Rome is an amazing place with many history behind every runes and structures. Every attractions is always packed with tons of tourists. You will never missed it as everyone will surround it like there is a circus performance on the spot. It is not easy to travel by walking around as you may lost your way easily. However, the Metro is a very good guide for us.
To start off, visit COLOSSEO, the place where gladiators fight with their life on the line in order to survive. The place grand from the outside (especially at night). There are mazes right at the center of the stage. I was told that the mazes used to guide the deadly animals to the stage above.
From the Colosseo Metro, you may take to Ottaviano Metro and take a 5 mins walk to where MUSEI VATICANI is located. This is a place where Catholics know better than me. It is like their headquarter. I can say this is the biggest and most amazing catholic church I've ever seen.
From the Ottaviano Metro, take to Flaminio Metro and walk via del Corso. Along this road you may shop and eat wherever you want. But don't forget to visit Piazza Miganelli, Fontana di Trevi (hold a coin with your right hand, back face the fountain, make a wish and throw it over your right shoulder) and Pantheon (a hole on the ceiling and was said that no water will come in even when it rains).
